How much damage would a 500 Nitro do?

A lot.
Elephant Gun and Big Game hunting rounds should be less potent than heavy machinegun rounds, but definitely stronger than regular rifle rounds or shotgun slugs.
So I would recommend looking about halfway between a high powered rifle round and a heavy machinegun round and assigning a representative number.

If my Recon books weren't boxed up I could give you a number for Recon.
Most SDC Games I'd give it a 6D6 or a 1D4x10.
